The effect of vitamin K1 and K2
Vitamin K and blood clotting
Vitamin K – the “K” stands for coagulation – has long been known as a central building block of blood clotting. It activates certain proteins that control this process in the body. If vitamin K is lacking, these clotting proteins remain inactive, meaning the blood no longer clots normally.
Blood-thinning medications are also based on exactly this principle: they specifically inhibit the effect of vitamin K, slowing down clotting.
The vitamin K-dependent proteins involved in blood clotting include: prothrombin, proconvertin, factor IX, factor X and plasma protein C & S
Vitamin K and calcium metabolism
Vitamin K works not only in blood clotting, but also supports the maintenance of healthy bones:
More recently, research has discovered additional functions of vitamin K – especially in connection with calcium balance. Special vitamin K-dependent proteins play a role here, occurring, for example, in bones and blood vessels. Many of these proteins were identified through experimental studies in which they were specifically deactivated.
These include, among others: osteocalcin, matrix Gla protein, Gla protein and protein S
Osteocalcin is especially important, as it is produced in bone and tooth tissue, where it binds calcium and hydroxyapatite – a key step in building stable structures.
Today, around 20 additional vitamin K-dependent proteins are known, and some of their exact functions are still being researched. For example, the protein Gas6 has been attributed an influence on the immune system.

Why both vitamin K1 and vitamin K2?
In principle, both forms – vitamin K1 and K2 – are able to activate K-dependent proteins in the body. Nevertheless, there are differences in how they work and how they are distributed.
Vitamin K2 in the MK-7 form remains in the blood significantly longer than K1 and is therefore considered around four times more effective.
Conversely, vitamin K1 can be enzymatically converted in the body (cleavage of the side chain) into other K vitamins, such as K2-MK4. This became clear because K2-MK-4 was detected in many tissues – even when only vitamin K1 was supplied. In other words, vitamin K1 serves in the body as a starting substance for MK-4, which can perform important functions locally.
For this reason, it makes sense to take both vitamins – K1 and K2. They complement each other in their effects and together help supply various tissues optimally with active vitamin K.
Is vitamin K necessary for vitamin D to work?
Yes and no. Vitamin K does play an important role in certain effects of vitamin D. For example, vitamin D stimulates the formation of osteocalcin – a protein that is important for bone metabolism. However, for osteocalcin to be effective, it must be activated by vitamin K. Without this activation, it cannot fully perform its function in the body.
At the same time, current studies show no additional benefit when vitamin K is taken in addition to vitamin D – provided vitamin K levels are within the normal range.
Conversely, this means: if there is a vitamin K deficiency, it could impair the activation of osteocalcin and therefore certain effects of vitamin D.
