Vitamin K and Blood Clotting

Vitamin K—the “K” stands forcoagulation —has long been recognized as a key component of blood clotting. It activates certain proteins that regulate this process in the body. Without vitamin K, these clotting proteins remain inactive, preventing the blood from clotting normally.

Blood-thinning medications work on exactly this principle: they specifically inhibit the action of vitamin K, thereby slowing down blood clotting.

Vitamin K not only plays a role in blood clotting, but also helps maintain healthy bones:

More recently, research has uncovered additional functions of vitamin K—particularly in relation to calcium metabolism. Specific vitamin K-dependent proteins, found for example in bones and blood vessels, play a role in this process. Many of these proteins were identified through experimental studies in which they were specifically inactivated.

These include, among others: osteocalcin, matrix GLA protein, GLA protein, and protein S

Osteocalcin is particularly important because it is produced in bone and dental tissue, where it binds calcium and hydroxyapatite—a key step in the formation of stable structures.

To date, around 20 additional vitamin K-dependent proteins have been identified, some of whose exact functions are still being studied. For example, the protein Gas6 has been found to influence the immune system.

Why both vitamin K1 and vitamin K2?

In general, both forms—vitamin K1 and K2—are capable of activating K-dependent proteins in the body. However, there are differences in how they work and where they are distributed.

Vitamin K2 in the form of MK-7 remains in the bloodstream significantly longer than K1 and is therefore considered to be about four times more effective.

Conversely, vitamin K1 can alsobe converted into other K vitamins, such as K2-MK4, through enzymatic conversion (cleavage of the side chain) within the body. This was demonstrated by the fact that K2-MK4 was detected in many tissues—even when only vitamin K1 was administered. In other words, vitamin K1 serves as a precursor for MK-4 in the body, which can perform important functions locally.

For this reason, it makes sense to take both vitamins—K1 and K2. Their effects complement each other, and together they help ensure that various tissues receive an optimal supply of active vitamin K.

Is vitamin K necessary for vitamin D to be effective?

Yes and no. Vitamin K does indeed play an important role in certain effects of vitamin D. For example, vitamin D stimulates the production of osteocalcin—a protein that is important for bone metabolism. However, for osteocalcin to be effective, it must be activated by vitamin K. Without this activation, it cannot fully perform its function in the body.

At the same time, recent studies show no additional benefit from taking vitamin K in addition to vitamin D—provided that vitamin K levels are within the normal range.

Conversely, this means that a vitamin K deficiency could impair the activation of osteocalcin and, consequently, certain effects of vitamin D.
